Liposomal Delivery Systems: Why Encapsulation Transforms Supplement Bioavailability

The Bioavailability Problem

The majority of oral supplements face a fundamental barrier: the gastrointestinal tract. Conventional capsules and tablets must survive stomach acid, intestinal enzymes, and the first-pass metabolic effect of the liver before any meaningful amount reaches systemic circulation. For hydrophilic compounds like Vitamin C and Glutathione, absorption often plateaus at doses above 200mg due to saturable intestinal transporters (Levine et al., 1996, Annals of Internal Medicine).

How Liposomes Work

Liposomes are spherical phospholipid bilayer vesicles — identical in structure to human cell membranes. When a nutrient is encapsulated within this bilayer, it bypasses the conventional absorption pathway. Instead of competing for intestinal transporters, liposomal particles are absorbed via endocytosis and directly enter the lymphatic system, achieving peak plasma concentrations significantly faster and at far lower doses.

A landmark comparison study by Bhattacharyya et al. (2009) demonstrated that liposomal Vitamin C achieved plasma concentrations 1.77× higher than equivalent doses of oral ascorbic acid at the same dose. More recent data from Gopi & Balakrishnan (2017, International Journal of Pharmaceutics) confirmed that encapsulation efficiency above 85% was the key threshold for clinical superiority.

Clinical Implications

For compounds like Glutathione — which is almost entirely degraded in the gut before absorption — liposomal delivery is not merely preferable, it is often the only clinically viable oral route. Research by Richie et al. (2015, European Journal of Nutrition) confirmed that 500mg/day of liposomal glutathione over four weeks significantly increased whole-blood glutathione levels, natural killer cell activity, and reduced oxidative stress markers. Standard oral glutathione failed to produce equivalent results.

AXEL TerraMatrix™ Application

All AXEL Lipo® formulations are designed with phosphatidylcholine-dominant bilayers (≥80% PC) to ensure structural integrity and targeted intracellular delivery. This is why clinical dosing protocols built on Lipo® products can achieve therapeutic outcomes at lower absolute doses — the nutrient reaches the cell.
